We don't know for sure Tohru is going to be in the stage play.

With the news that it's going to be an all-male troupe I wouldn't be surprised if maybe the stage play is set shortly before the manga series begins. That way it removes the three main female characters from the series. It also of course spoiler[could explain why Akito, should she be in it, be played by a guy since at that point virtually all the major characters would naturally assume that she is a guy. Also the two female zodiac members are, for the most part, not integral to the story so they don't have to be in it either.]
