Was it ever explained in the manga why Mista dislikes the number 4 so much?

Because in Japanese culture, 4 is an unlucky number associated with death.

I'm just gonna guess that something traumatizing happened to him when he was 4 and he doesn't want to think about that time, and just leave it at that.

Yes.
In his early childhood, one of Mista's neighbors adopted a kitten out of four, and later learned that kitten clawed one of said neighbor's eyes out. This resulted in Mista's fear of the number 4.
In other words it was very disturbing for a child.

Yes.

I'll admit I'm a little hazy on Part 5 since I was kinda quick to dismiss it as a dull return to formulaic villain of the week stuff after Part 4 got super creative with most enemy stands, but I don't remember that little tidbit. That wasn't from the Fugo LN, was it?
