They're basically joke chapters of the regular manga. They have silly stories that aren't important to the overall storyline of the series.

On the subject of FMA...

Is the anime "permanently" ended? That is to say, there is no prospect of a continuation in the future?

Yes, unless they announce more, the anime has official ended with the movie.

I recently heard from a friend of mine, who has read all of the FMA manga, that the ending to the manga is completely different from the ending of the anime. He even said that the story changes completely around the middle or so (around ep. 30).

They are pretty much two different series (different plot, characters, villian, etc.). Except that the manga isn't finished yet, volume 13 was just released in Japan last week (a new chapter comes out once a month in Shonen Gangan).

On gaidens, they're just extra stories. There are some that aren't humorous, like the "The Blind Alchemist" and "His Battle Field Once More" (gives some good insight into Hughes and Mustang). Key difference in the manga, Hughes participated in Ishbal. Wink
