So is Kira's assistant always in the form of Misa? I mean, most convenient way of letting Aya Hirano reprise her role.

That's right, Kira's assistant in Death Note often appears as Misa Amane, which makes it convenient for Aya Hirano to reprise her role. Misa, a staunch supporter of Kira, is a central character with ties to Light Yagami and often acts as his accomplice throughout the series.
